there are many instances of increases in genetic material being harmful. down syndrome is an obvious one. but consider- amoeba have much much more genetic material than humans. get over the notion that more is better; it isn't

there are any number of instances of gene duplication followed by mutation leading to increase of fitness- we see in three colours because an opsin gene in an ancestor was duplicated and through an identified mutation on the duplicate a different frequency was sensed. the gene remains. we don't survive by smell any more- we have multiple disabled odor rceptor genes.

it's a beneficial mutation. will you people stop moving the goalposts. it is a random mutation in a globin gene that if it affect both copies of the gene you get SC anaemia which is harmful and if you get one copy you get SC trait. making you less suceptible to malaria and hence confers an advantage. EV would predice that this would be selcted for in areas where malaria is a major threat to survival. guess what- that's exactly what is found.
